Circulating Thrombospondin-1 and Endothelin-1 Levels Tend to Decline with Increasing Obesity Severity in Women: Evidence from a Pilot, Cross-Sectional Study
Marta Greco et al.
Thrombospondin-1 (TSP1) is a multimeric glycoprotein implicated in metabolic, thrombotic, and inflammatory pathways and associated with adipose tissue dysfunction and insulin resistance, although its link to obesity severity remains uncertain. Endothelin-1 (ET1), a regulator of vascular homeostasis, may also contribute to obesity-related cardiometabolic risk with possible sex-specific effects.
This study evaluated circulating TSP1 and ET1 in nondiabetic obese women, analyzing associations with metabolic and inflammatory parameters and differences according to obesity severity and disease risk.
Protein quantification was performed using the automated Ella™ microfluidic ELISA platform, which enables high-sensitivity detection with minimal sample requirement. The system operates through single-use cartridges pre-loaded with monoclonal capture antibodies specific to TSP1 and ET1, allowing serum samples to flow through microfluidic channels where the analytes are selectively bound and quantified. Each measurement was performed in triplicate, providing increased analytical precision and reliability.
